From the FAQs (little red link at the top right)
What are the differences between open worlds and realistic worlds?
Open worlds (odd numbered) have no political boundaries, so you can fly anywhere in the world regardless of your home base. You can lease up to 40% of the gates at any one airport (75% if it's a hub); building a terminal removes this limit.
Realistic worlds (even numbered) contain real-world political restrictions and additional route taxes and fees. For example, if you are based in country A, you can fly between country A and country B or country A and country C but not between country B and country C, unless an open skies agreement specifically allows for the latter (e.g. the European Union). Certain country pairs have additional flight prohibitions (e.g. between the U.S. and North Korea). You can lease up to 5 gates at any one airport (15 if it's a hub); building a terminal removes this limit.

To be simple:
In open worlds, Airlines can lease gates and operate from any country they want. For example, your HQ is in ATL, Then you leased a gate at PEK, You can operate routes from there like this:
PEK-BKK
Notice that you can operate almost every part of the world in your freedom, but watch out. Big airlines are a major threat in open worlds since they might expand and annihilate your airline.
In Realistic Worlds, You can only operate 1 country on where your HQ is unless there is an open skies agreement (Eg. EU open skies) For Example, Your HQ is in DXB, You can only operate flights from the UAE only like:
DXB-DOH
AUH-DOH
and other more.
Realistic worlds are my favorite, Big airlines have less chances to annihilate your airline, So I recommend Realistic Worlds as the ones you want to play in unless you want a bigger challenge.
Have A Nice Day

i was only able to fly from amsterdam to those airports, but not from Glasgow-Luton/Stuttgart, or Luton-Stuttgart
