the route has been recalculated by server or you have changed something on it eg IFS or you opened a hub at an airport

As advised above, It's a route re-calculation symbol & can happen for a variety of reasons. It may mean a competitor has just adjusted their prices against yours & your market share is being recalculated. Or, if you've just opened a hub, it will be re-adjusting capacity & the mix of green & blue (connecting) passengers. It will clear in time (depending on the processing backlog) or you can go into the route (click "manage") & then just click the "save" button on the main route screen. It will force the recalculation to complete (your daily profit figure will probably change) & the icon will be gone next time you're in your routes menu.
