United has ended HKG-SIN as of November, 2017; replaced by the non-stop LAX-SIN flight.

UPDATE :
- moved the list to Google Docs
- added 2 features, respective flight numbers and aircraft information !!!
- added new airlines (such as MIAT Mongolian, Air Namibia, Mauritania Airlines, and many more)
- huge updates to Qantas, Ethiopian, Rwandair, Tunisair, Kenya Airways, Emirates, COPA and many more!

Uzbekistan Airways have 5th Freedom flights from SIN-KUL, as a tag on flight from TAS (I think it’s HY552/553).
Air Mauritius also have flights from SIN-KUL. Both are bookable on their own.
Added Air Mauritius to the list, can't believe I forgot about that. Thanks. As for Uzbekistan Airways, the route is unbookable on their own website and other 3rd party travel agencies, so it's pretty debatable.
I think the South African Airlines flight from JNB-HLE stops in WDH, but I’m not sure if you can book that individually. I’m also not 100% sure about whether or not it counts as a fifth freedom flight because St Helena isn’t a country, but it’s definitely not part of a South Africa of Namibia, so...

For St. Helena, the stop in Windhoek is for refuelling purposes, (here for source), hence, not a fifth freedom flight. As for Air Malta flights, Malta is a member of the EU, and EU operates an open sky policy, I wouldn't count that as fifth-freedom. Thanks anyway for the suggestions.

JY also operate the route, but that might not be 5th Freedom either, because although JY are based in the Turks and Caicos Islands, not the British Virgin Islands, they’re still both British overseas territories. Who knows?
I’m pretty sure JY621/622 DOM-SLU counts though.

What about MD18/19 RUN-CAN? While it does have its own Air Madagascar flight numbers, all the OTAs I checked only sell it as UU818 operated by Air Madagascar. However, it does show up on MD’s website, I just can’t get it to work well enough to actually search for a flight on my device.
There’s also a few LIAT flights (I think LIAT is only based in ANU, BGI & POS):

As for Ethiopian, I've updated the LAX routing, but the European flights, to my knowledge, aren't fifth freedom flights.

The plane makes a round-trip only stopping to let people on and off, for refuelling, and a quick interior clean at BGI, and keeps the same flight number throughout, so arguably it's a MAN-MAN flight

Hi, thanks for that, however, TAB-BGI isn't bookable on their website.
The LATAM flight is LA 896/897, operated by an A320. LA897 goes PUQ-RGL-MPN, while LA 896 continues to Santiago MPN-RGL-PUQ-SCL. According to the Falkland Islands travel specialists wbsite, “ On the second Saturday of each month, you can board the flight at Rio Gallegos in Argentina. On the third Saturday of each month you can disembark in Rio Gallegos.” It is bookable by itself on their website and on OTAs (although really hard to find any dates!) I just don’t know if LATAM Chile and LATAM Argentina are legally separated enough for it to count. They use different IATA codes (LATAM Chile uses LA but LATAM Argentina uses 4M) and Wikipedia describes them as different arilens, but are both in the LATAM Airline Group, which also includes LATAM Brasil, Comobia, Ecuador, Perú etc, so my guess is it counts.
Separately, there is also what is possibly the coolest fifth freedom flight in the world: Macau - Koror, Palau, operated by Lanmei Airlines, a Cambodian LCC on an A319. The flight numbers a LQ9508/9509.
Thanks again, however, I wouldn't count the Lanmei Airlines flight a fifth freedom, simply because I can't book it, heck I don't seem to be able to book a flight to Palau on their website. As for the LATAM one, I've added it to the list.

Hainan Airlines operate an once weekly PEK-EDI-DUB-PEK as well as another once weekly PEK-DUB-EDI-PEK in a triangle routing format with EDI-DUB bookable.
