Where/How does AE get the numbers for passenger demand for different routes?

No. The demand from A to B is not based on real life figures. What if the route doesn't exist in real life? You can't just go to a website that shows demand form everywhere to everywhere :P

We take the real-world total amount of passengers listed at each airport in the game and put them into a database. The game runs a script to determine the demand on each route, based upon this information. This is not perfect, but it is nigh on impossible to find or manufacture demand for every single city pair possible.

The script needs tweaking - for now, it doesn't take into account a lot of things. For instance, if there's an airport that has two million annual passengers, but in real life all of those passengers are on a single route, the AE script will underestimate the demand on that given route and overestimate it on all others. It also overlooks other important factors like poor ground communication to certain regions and certain political issues. Its not perfect but it is a work in progress though!
